You will need to initialize CartonSuffix by running an Update query.
UPDATE MyTable
SET CartonSuffix = 0
WHERE CartonSuffix IS NULL

While selecting both fields CartonNo and cartonSuffix by holding down the shift and then clicking on Primary key both fields become Primary key, but while saving the table i get an error message "Index or Primary key cannot contain a null value".
