I'm getting the same issue with one of my clients today. It's obvious between any 2 PCs.
Network folder has full access for both users. I've created a new blank database file & as you've found if one PC has it open, the other PC can't. Both PCS are opening the file as 'shared' rather than exclusive. 
PC1: Windows 10 Pro, Access 365, v2111 build 14701.20226
PC2: Windows 10 Pro, Access 365, v2111 build 14701.20248
I think there was an update overnight, but now only one user at a time is able to use the database.
I haven't found anything on any other forums about this, but I'm expecting my phone to start blowing up as other companies get new version of Office - thanks MS, Merry Christmas :-(
