I seem to recall a bug several versions ago regarding ascending /
descending. If you created a descending index on a field, then tried to
make it the Primary Key, Access would crash. This implies to me that Access
(JET / ACE) does in fact build a descending index different from an
ascending one. If this is true, then yes, it does make a difference.

I was in another forum and a question came up as to when one should index
using descending instead of Ascending, and does it make any difference or is
